A novel genotype of MVA that efficiently replicates in single cell suspensions

چکیده
Background Vectored vaccines based on modified vaccinia Ankara (MVA) may lead to new treatment options against infectious diseases and certain cancers. MVA is highly attenuated and requires avian cells for production. We established avian continuous cell lines (including CR and related CR.pIX) and adapted these cells to proliferation in single-cell suspension in a chemically defined medium [1,2]. Replication of several viruses was efficient in CR suspension cultures [3,4] but yields for MVA were low. We suspected that cell-to-cell spread may be an important mechanism for MVA replication in agitated suspension cultures and developed a production medium that is added at the time of infection to induce cell aggregates [2]. MVA (and other host-restricted poxviruses) replicate to very high titers with this robust and fully scalable cultivation protocol but further improvement may facilitate production for large vaccine programs. We now describe a novel genotype of MVA that replicates with high efficiency in single-cell suspensions without aggregate induction.

Prognostic Significance of MMP2 and MMP9 Functional Promoter Single Nucleotide Polymorphisms in Head and Neck Squamous Cell Carcinoma
Objective(s) Matrix metalloproteinases comprise a family of enzyme that is able to degrade components of extra cellular matrix. There are single nucleotide polymorphisms in the promoter regions of several genes with ability to influence cancer susceptibility. The aim of this study was to analyses association between MMP2 and MMP9 promoter polymorphisms and head and neck squamous cell carcinoma...
متن کاملO-36: Genome Haplotyping and Detection of Meiotic Homologous Recombination Sites in Single Cells, A Generic Method for Preimplantation Genetic Diagnosis
Background: Haplotyping is invaluable not only to identify genetic variants underlying a disease or trait, but also to study evolution and population history as well as meiotic and mitotic recombination processes. Current genome-wide haplotyping methods rely on genomic DNA that is extracted from a large number of cells. Thus far random allele drop out and preferential amplification artifacts of...

Background: Silica nanoparticles could enter the living animals and humans through inhalation, ingestion, injection and skin, exerting potentially adverse effects on health and environment. The aim of this experimental laboratory study was to evaluate toxicities of silica nanoparticles in the form of wire, rod and sphere with different sizes (20, 50 and 100 nm) on rat pulmonary cells after 6 an...
متن کاملElements in the Development of a Production Process for Modified Vaccinia Virus Ankara
The production of several viral vaccines depends on chicken embryo fibroblasts or embryonated chicken eggs. To replace this logistically demanding substrate, we created continuous anatine suspension cell lines (CR and CR.pIX), developed chemically-defined media, and established production processes for different vaccine viruses. One of the processes investigated in greater detail was developed ...
متن کاملI-38: Chromosome Instability in The Cleavage Stage Embryo
Recently, we demonstrated chromosome instability (CIN) in human cleavage stage embryogenesis following in vitro fertilization (IVF). CIN not necessarily undermines normal human development (i.e. when remaining normal diploid blastomeres develop the embryo proper), however it can spark a spectrum of conditions, including loss of conception, genetic disease and genetic variation development. To s...
